High molecular weight phthalate esters demonstrate no skin or eye irritation in rabbits and low acute toxicity by oral, inhalation, and dermal routes; Repeated dose studies produce effects including increase in peroxisomes with increases in other parameters associated with peroxisome proliferation (liver hypertrophy and increased peroxisomal enzymes), adverse effects to male and female reproductive systems, and teratogenic effects in several studies (mostly at or above doses maternally toxic); Very limited evidence for mutagenicity; [EPA ChAMP] Not irritating to skin or eyes of rabbits; A 21-day oral study of rats demonstrated evidence of peroxisome proliferation at 10% in the diet; [IUCLID] See "PHTHALATE ESTERS."
